Cyril Cook- Barrick lab: A study of how bivalency and disorder affect binding

 

Notch signaling is a conserved juxtacrine signaling mechanism that affects gene expression in metazoans, inducing cell fates such as differentiation, homeostasis, and apoptosis.  The notch transcription activation complex is ternary and comprises the transcription factor CSL and two co-activators, the intrinsically disordered MAML and the Notch Intracellular Domain (NICD) of the membrane-bound Notch receptor.  CSL is bound by two domains in the NICD, RAM and ANK, and these domains are connected by an intrinsically disordered linker.  Previous work in the lab shows tight binding between RAM and CSL, yet RAM is not required in the minimal activation complex needed for notch signaling activity.  Since all binary interactions between ANK, CSL, and MAML are weak compared to the binding between RAM and CSL, we are investigating the role of RAM in promoting the cooperativity of formation for the notch transcription activation complex.
